Effect of Pepsinization on the Immunohistochemical Staining of Paraffin-Embedded Tissues


Abstract
With liver, thyroid, pancreas, cervix tissues we carried the immunohistochemical ABC method in which Laminin was used for 1st antibody. The course of that method, we investigated the optimal treatment time and temperature conditions of pepsin. And then, the following results were obtained.
a. When materials were not treated with pepsin, all cases showed negative responses.
b. When materials were¢¥ treated with pepsin at 37¡ÆC for 3hrs, all cases showed strong positive responses.
c. When materials were treated with pepsin at 47?C for 2hrs, all cases showed week (+) responses.
d. When materials were treated with pepsin at 57?C for l hr., all cases showed considerably weak (+, -) responses.
In view of the results so far achieved, we concluded that best responses was obtained when pepsin was treated at VC.
